Isolation and Characterization of Phb Producing Micro-organisms Isolated from Root Nodules of Leguminous Plants

چکیده
A number of the micro-organisms are capable of producing polyhydroxyalkanoates (PHA’s) as storage food reserve under unbalanced growth conditions. The mechanical properties of PHA’s are similar to that of synthetic plastics like polyethylene, polypropylene etc. The advantage of using PHA’s over synthetic plastic is that PHA’s are completely mineralized into carbon dioxide and water through the action of various microorganisms. Polyhydroxybutyrate (PHB), Polyhydroxybutyratecohydroxyvalerate (PHBV) are the best known representatives of PHA family. These representatives of PHA can be used to produce environment friendly alternative to the synthetic polymers which pose a number of environmental problems. PHB are produced intracellularly by various organisms such as Bacillus megaterium, Ralstonia eutrophus, Cupriavidus necator, Rhizobium spp., Azotobacter spp., Pseudomonas spp., etc under physiological stress conditions. These bacteria can accumulate up to 60-80% of their weight as PHB under limiting nitrogen substrate and in the presence of an abundant source of carbon (Anderson and Dawes, 1990). In last few years, various attempts have been made to use PHB for the production of biodegradable plastics (bioplastic) (Mercan, 2002; Mukherjee et al., 2011). The major limitation in the use of biodegradable plastic is their high cost as compared to the synthetic plastic. It is well established that the cost of production of any microbial product can be reduced significantly if these micro-organisms are harvested on low cost media having agro-industrial byproducts since media is the main component that add to cost of production (Dhingra, 2012; Goyal and Dhingra 2011).

Isolation and Characterization of Agrobacterium rhizogenes from the Root Nodules of Some Leguminous Plants
A total of 57 colonies of Agrobacterium species was isolated from the root nodules of five different leguminous plant namely Pisum sativum, Sesbania rostrata, Vigna mungo, V. radita and V. ungiculata by using yeast extract mannitol agar (YEMA). All the isolated colonies were belonged to Agrobacterium species. The isolated bacteria were characterized and identified as Agrobacterium rhizogenes ba...

Polyhydroxybutyrate (PHB) is a polyhydroxy alkanote (PHA) which produces in ralstonia eutropha by three enzymes, including Beta-ketothiolase, NADPH-dependent acetoacetyl-CoA reductase and PHB synthase. These three enzymes are encoded by phbA, phbB and phbC respectively. There is considerable interest in Polyhidroxybutirate, since it can be used as biodegradable plastic, which under optimal cond...
متن کاملIsolation, Screening and Characterization of Hyaluronidase Producing Bacteria
Hyaluronidase has a panoramic use in biotechnology processes and therapy due to its therapeutic, pathophysiological, physiological and biological importance. Since much of the preparations of hyaluronidases are from animal source (bovine and ovine testicular sources) with limited sources of microbial origin, that prompted the authors to screen and isolate a new promising bacterial strain ...
متن کاملCharacterization of Agrobacterium rhizogenes Isolated from the Nodules of Some Leguminous Plants
Agrobacterium strains were isolated from five different leguminous plants viz., Pisum sativum, Sesbania rostrata, Vigna mungo, V. radita and V. ungiculata. The isolated bacteria was characterized and identified as Agrobacterium rhizogenes based on their morphological, biochemical, cultural and pathogenicity tests. Antibiogram and protein profile of isolates were also studied by disc diffusion a...
متن کاملIsolation and Characterization of Melanin Producing Pseudomonas stutzeri Strain UIS2 in the Presence of l-tyrosine and Survey of Biological Properties of Its Melanin
Background: Melanin is a negative charge hydrophobic complex pigment. Melanin is produced naturally in bacteria to protect them against UV, free radicals and environmental stresses. Pigment production in bacteria has more advantages than other biosources due to its rapid growth, higher efficiency and easier extraction. The aim of this study was the isolation, biochemical and molecular identific...
